< Abstract >
In
spite of being rare, gallbladder cancer (GBC) is extremely aggressive and
deadly malignancy of the biliary tract with striking geographical, race, and
gender-related differences in its incidence. It is associated with late diagnosis,
unsatisfactory treatment, and poor prognosis. Apart from gallstone, and female
gender being the documented non-modifiable risk factor for GBC, the
pathobiological mechanisms underlying the phenotypic expression of GBC appear
to be rather complex. Extensive epidemiological studies have demonstrated that
genetic variants, mainly single nucleotide polymorphisms (SNPs), are likely to
modulate the consequence of environmental risk factors (dietary carcinogens
exposure such as tobacco, alcohol, etc) through modifying functions of various
biological pathways concerned in gallbladder carcinogenesis. Detection of these
risk sets of genetic variants will facilitate in determining individuals at
higher risk for developing GBC. Therefore, we performed case-control study
including 400 GBC and 250 healthy controls (HC) and investigated SNP genotypes
in several genes belonging to different pathway such as tumor suppresser gene
(DCC), gallbladder contraction (ADR), apoptosis (DR4, FAS, FASL), estrogen
metabolism and steroid hormone biosynthesis (CYP17). Genotyping was performed
by PCR-RFLP or taqman assay. Statistical analysis was performed by using SPSS
ver16. Functional prediction of these variants was carried out using
Bioinformatics tools. We found that these variants either alone or in
combination (haplotype) may have significant
effect on GBC susceptibility that may be explored as a useful marker for GBC
susceptibility. However, additional well-designed studies with larger sample
size and focusing on different ethnicities are required to further validate the
results.
