< Identification and Biosynthesis of New Secondary Metabolites from the Thermophilic Bacterium Thermosporothrix hazakensis >

Microbial natural products have been excellent sources for drug discovery and development over the past three decades, but the isolation of known compounds is a common challenge that hinders the discovery of novel bioactive compounds. Because of this problem, new biological sources such as thermophile are needed for the discovery of novel natural products. Thermophilic bacterium Thermosporothrix hazakensis SK20-1T having unique characteristic of multiple exospores per mother cell by budding in branched aerial mycelia was considered to produce unprecedented metabolites.
The genome analysis of SK20-1T revealed the presence of 23 gene loci related to secondary metabolism. Further bioinformatics analysis with antiSMASH software provides the genetic potential for production of secondary metabolites including four-thiazoline-containing compound, lipohexapeptide, and cinnamycin-like lantipeptide. In addition, we explored secondary metabolites from cultures of SK20-1T to examine production of compounds expected by the bioinformatics analysis. Through the chemical study, we discovered five new compounds including two acyloins, two indole alkaloids, and a polypropionate.
In case of new two acyloins, genome-based screening and biochemical studies identified the thiamine diphosphate (TPP)-dependent enzyme Thzk0150, which is involved in the formation of acyloin. Through extensive analysis of the Thzk0150-catalyzed reaction products, we proposed a putative reaction mechanism involving two substrates: 4-methyl-2-oxovalerate as an acyl donor and phenyl pyruvate as an acyl acceptor.
