Aging in muscle is associated with a decrease in mass and strength. Human loses up to 40% muscle at the age of 65. To fully characterize molecular network of aging in skeletal muscle, we performed next-generation sequencing to assess genome-wide expression profiling of microRNA and mRNA as well as DNA methylation, using mouse gastrocnemius muscle at two different ages (6-month-old versus 24-month-old). We found 34 age-regulated miRNAs and 16 novel miRNAs in muscle. Genes regulated either by epigenetic or transcriptional control contribute to increase of inflammatory response, stress response, and growth inhibition, while decreasing energy metabolism and muscle development. Age-associated miRNAs might contribute to aging through the control of glucose metabolism, ion homeostasis, and transcription regulation. By profiling proteomic changes during muscle aging, we found several protein markers useful for diagnostics and characterized the plausible deteriorate mechanisms by the proteins. These events together drive the decrease in myoblast differentiation and the increase in atrophy, leading to muscle aging. Further, we investigate the functions of candidate genes and regulatory networks in the physiological levels. Also we screen drug candidates that enhance muscle regeneration by using our unique assay system. We have selected several positive candidates in animal test. One candidate are now under a clinical study to develop future therapeutics to overcome muscle aging.
